Application of dsRNA VP15-WSSV by Oral Vaccination to Increase Survival Rate and Response Immunes of Tiger Shrimp Penaeus monodon

Background: RNA interference (RNAi) has recently introduced as a powerful technique for specific gene silencing in antiviral therapy controlling shrimp diseases. The present study was conducted to assess the effect of dsRNA VP15-WSSV vaccine by oral administration on tiger survival rate and response immunes. Methods: A encoding VP15 isolated from infected WSSV cloned into L4440 vector with T7 promoter. For mass production, pT7-VP15 construct transformed bacteria. bacteria were inactivated using heat-killed method immersion 108 cells mixed 0.02 g commercial feed application. fed by: (A) containing coated fish oil, (B) without oil (C) oil. challenged intramuscular injection after vaccination two weeks. (SR) challenge test counted daily, while THC proPO observed before, then 1st, 3rd 5th days post-challenge (dpc). Result: all treatments tended decrease until dpc, 4th dpc vaccinated started exhibit higher SR compared both treatments. had significantly (P less than 0.05) control treatments, but not greater activity. results indicated that application suggested increasing immune shrimp.
